Immunocytochemical analysis of human cells after 6 days of differentiation. (A) shows strong homogeneous staining for desmin of the cells. (B) The differential capacity of the multinucleated cells was demonstrated by myogenin-positive staining of the majority of the nuclei. (C) Strong homogeneous staining of differentiated cells with an antibody against titin. (D) The vast majority of differentiated cells showed strong staining with the antibody against MyHC-embryonic and (E) many myotubes expressed neonatal MyHC. (F-H) expression of fast and slow MyHC was revealed in a subset of fused cells using antibodies against these isoforms. Positive staining of differentiated cells with antibodies against slow (I) and fast (J) MyBPC, respectively. The differentiated cells showed weak positive immunoreactivity with the antibody against alpha-cardiac actin (K) but stronger positive staining with alpha-skeletal actin (L).
